MEN 2 (Multiple Endocrine Neoplasia Type 2)
An inherited syndrome causing endocrine tumors including MTC — an absolute contraindication to GLP-1 therapy.
MEN 2 is a hereditary syndrome, driven by RET gene mutations, in which endocrine glands develop tumors — most consistently medullary thyroid carcinoma, often alongside adrenal pheochromocytomas and parathyroid disease. Because MTC in MEN 2 approaches certainty over a lifetime, carriers are managed intensively, sometimes with preventive thyroidectomy.
For weight treatment the implication is the same bright line as MTC itself: a personal or family MEN 2 diagnosis contraindicates semaglutide and tirzepatide entirely. If endocrine tumors cluster in your family history and no one has named a syndrome, mentioning that pattern at your consultation — and to your primary doctor — is worth doing regardless of weight medication plans.
